Pancreatic levels of Bcl-xL mRNA increase in cerulein pancreatitis. Total RNA was extracted from pancreatic tissue of rats and mice with fully developed cerulein (CR) pancreatitis (at 7 h), as well as from control animals. RNA was subjected to RT-PCR as described in Materials and methods, using primers presented in Table 1. The gene for acidic ribosomal protein P0 was used as a reference (“housekeeping”) gene. (A) Expression of the main Bcl-xL transcript (transcript variant 3 in the rat) was quantified by real time RT-PCR and normalized to that in control group. Values are means ± SE from 3 animals per group. (B) Expression of rat Bcl-xL transcript variant 1 was measured by semiquantitative RT-PCR Each lane represents an individual animal; shown are the results for 2 rats per group. Numbers to the right of the gel are DNA size markers in base-pairs.
