Rats were euthanized 30 min after one injection of methamphetamine (2 mg/kg, i.p.) or vehicle, and striatal homogenates were fractionated by differential centrifugation. S3 = cytosol, P1 = nuclei and large debris, P2 = crude synaptosomes, LP1 = synaptosome membranes, LP2 = synaptic vesicles, P3 = light membranes (non-synaptosome). The fractions and initial homogenates, containing equal amounts of protein, were loaded into SDS-PAGE gels and analyzed by western blot for Rab10 immunoreactivity. Results shown are the mean ± SEM of 4 samples, expressed as the ratio of Rab10 immunoreactivity in each fraction over Rab10 immunoreactivity in the initial homogenate. *p < 0.05 (unpaired Student’s t-test).
