NLRP3, apoptosis-associated speck-like protein containing CARD (ASC), and caspase 1 are dispensable for tumor necrosis factor α (TNF-α) and interleukin 6 (IL-6), which are secreted in a canonical pathway. (A, B) Tnfa and Il6 mRNA expression in bone marrow–derived macrophages (BMDMs) from wild-type (WT; C57BL/6), NLRP3-/-, ASC-/-, or Casp1-/- stimulated by Porphyromonas gingivalis strains 381 or DPG3 at a multiplicity of infection of 100 for 3 h. Total RNA from each group was extracted, reverse transcribed, and quantified by quantitative reverse transcription polymerase chain reaction.
