In contrast to cholesterol-enriched wild-type macrophages, cholesterol-enriched ABCG1^−/− macrophages deposit little cholesterol unless treated with TO9. ABCG1^−/− mouse bone marrow-derived macrophages were incubated for 4 days with either TO9 (5 μM), AcLDL (50 μg/ml), or AcLDL + TO9 before cultures were immunostained with anti-cholesterol microdomain MAb 58B1 (green fluorescence) and DAPI nuclear stain (blue fluorescence).
