Inhibiting ABCA1 with probucol in cholesterol-enriched wild-type macrophages blocks TO9-stimulated cholesterol deposition. Wild-type mouse bone marrow-derived macrophages were incubated for 4 days with either AcLDL (50 μg/ml), AcLDL + TO9 (5 μM), or AcLDL + TO9 + probucol (10 μM) before cultures were immunostained with anti-cholesterol microdomain MAb 58B1 (green fluorescence) and DAPI nuclear stain (blue fluorescence). Upper and lower rows are, respectively, the fluorescence and phase photomicrographs. Scale bar = 50 μm and applies to all.
