Ang-4 and LEC permeability. LECs were seeded to confluence in the upper chambers of transwells and treated with 0.4 µg/ml Angs or indicated concentrations of VEGF-C for 4 h. FITC-Dextran 40,000 (1 μg/ml) was added to each chamber for 1 h. Permeability of the EC monolayer was determined by measuring the fluorescence of the medium in the bottom chamber using a Fluoroskan Ascent with Ascent 2.6 software for acquisition. Graphs represent the mean fold change relative to control from 8–10 combined experiments. Statistical analysis was performed using 1-way ANOVA and Tukey’s HSD post hoc test.
