Effects of PreP overexpression and activity on mitochondrial and cerebral Aβ load in mAPP mice. (A) PreP activity for degrading Aβ. PreP-enriched mitochondria from APP/PreP mice degraded significant amount Aβ1-42 as shown by less Aβ immunoreactive band, compared with mitochondria from APP or APP/PE mice. The addition of anti-PreP antibody to mitochondrial PreP protein extracted from APP/PreP mitochondria completely blocked degradation of Aβ (lane 8). Lane 1 denotes biotin-Aβ (10 ng) as a positive control. (B–E) Aβ levels in synaptic mitochondrial fractions (B and C) and cerebral cortex and hippocampus (D and E) from the indicated Tg mice were measured by ELISA. N = 6–14 mice per group.
