Signaling properties of the WT and mutant hMC3Rs with NDP-MSH as the ligand. HEK293T cells were transiently transfected with WT and mutant constructs, and intracellular cAMP levels were measured by RIA after stimulation with different concentrations of NDP-MSH as described in Materials and methods. Data are mean ± SEM from triplicate measurements within one experiment. All experiments were performed as least three times independently.
