Repression of the 1.5 kb nit-6 promoter fragment by Gln. Strains were cultured in VM-nitrate medium overnight and then transferred to VM-Gln (20 mM glutamine) for the indicated amounts of time (0.25–3 hr). Northern and Western analysis were performed using total RNA and protein extracts isolated as described for Figure 2. The asterisk indicates a nonspecific background band. Strains are wild-type 74-OR23-IVA (WT), pccg-1_GFP, and ptcu-1_1.5. Treatments are the same as in Figure 2. The blot shown is representative of at least three experiments.
