MiR-587 confers resistance to 5-FU treatment in colon cancer cells in vitro. (a) miR-587 expression was increased 16–20-fold in HCT116 and GEO cells as compared with vector control, and ectopically expressing miR-587 as determined by quantitative real-time PCR. (b and c) miR-587- and vector-expressing cells were treated with increasing concentrations of 5-FU for 72 h. MTT assays (b) and DNA fragmentation assays (c) showed increased resistance to 5-FU-induced apoptosis in miR-587-expressing cells compared with the control cells. (d) Western blot analysis showed that cleaved PARP was lower in miR-587-expressing cells than vector control cells after 5-FU treatment (HCT116, 10 μg/ml; GEO, 20 μg/ml) for 72 h.
