Effect of RAD1901 on uterine tissue. Newly weaned female Sprague–Dawley rats were treated with vehicle, E2 (0.01 mg/kg), raloxifene (3 mg/kg), tamoxifen (1 mg/kg), RAD1901 alone (0.3 to 100 mg/kg), or RAD1901 (0.01 to 10 mg/kg) in combination with E2 (0.01 mg/kg) once daily for three consecutive days. (a) Twenty-four hours after the final dose, rats were killed and uterine wet weights were recorded in grams. (b) Epithelial height was quantified in tissue sections of the uterus. (c) Representative sections of Toluidine Blue O-stained uterine tissue at ×400 magnification. Arrows indicate the uterine epithelium. (d) Total RNA was extracted from uterine tissue and analyzed by quantitative RT-PCR for the level of complement C3 expression relative to the 18S ribosomal RNA housekeeping gene.
