Bottom-up quantitative membrane proteomics may serve as a hub that connects various structure and function technologies, to accelerate discovery of the structure–function mechanisms of signaling TM proteins for better therapeutics (Path 2). Breaking the barriers against direct proteomic analysis of functional-state membrane proteins, at high peptide reproducibility and coverage, is key to reaching this goal.
