Model suggesting roles for K14-dependent disulfide bonding toward the organization and dynamics of keratin IFs in skin keratinocytes. We propose that Cys-4 and Cys-40 and their dependent disulfide bonding make a significant contribution to regulating the early stages of filament assembly (e.g. nucleation and elongation), whereas Cys-367 and its dependent disulfide bonding play a significant role in the genesis and/or maintenance of perinuclear IF networks as part of the keratin filament cycle (15). Pink and blue shades represent regions of relatively oxidizing and reducing environment within the cell, respectively, based on evidence from the literature (23).
