Matrix compliance regulates cytokinesis in HDFs. Substrate compliance affects cytokinesis in HDFs. Mitotic HDFs were collected by the shake-off method and replated onto fibronectin-coated hard and soft hydrogels in DMEM with 10% FBS. Cells were incubated for 3 h or overnight at 37°C and then fixed and stained to visualize α-tubulin (red) and nuclei (blue). (A,B) Plotted are the mean percentages of binucleated cells±s.e.m. on hard and soft substrates after either a 3 h (A) or overnight (B) incubation. One hundred cells were counted per condition for each of three independent experiments (**P<0.005). (C) Representative images of cells on hard and soft substrates are shown. Inset: magnified view of nuclei. Bar, 25 µm.
