We used FCS to investigate ROIs at various cell locations, both in the cytosol and in the nucleus but far from areas of heterochromatin. In this instance, Cerulean-CTA FP was used to image the cytosol and mCherry RFP was used to tag BZip protein domains; the tagged BZips were observed diffusing in the nucleus via FCS. Because BZip preferentially attaches to heterochromatin, we were able to choose ROIs far from heterochromatin by avoiding RFP congregation areas (bright red spots).
