Specific activity of glucose uptake in HEK293 cells stably expressing PfHT and GLUT1 shRNA at different lopinavir concentrations. Glucose uptake was determined by quantifying the amount of 2DOG accumulated in HEK293 cells and normalizing to time and total protein. The experiment was repeated in an identical background cell line that does not overexpress PfHT. Normalized specific activity of PfHT-overexpressing cells was adjusted for non-PfHT-specific glucose uptake by subtracting background uptake. Data are expressed as means ± SEM. IC₅₀ was calculated using nonlinear regression analysis.
