Values of the first-order rate constant for displacement kdisp were obtained from the data shown in Fig. 5B via nonlinear least-squares fitting with kobs = kdisp Ptot/(Aen + Ptot), where Ptot represents a total concentration of Egr-1 ZF-DBD, and Aen represents an apparent steady-state constant for the encounter complex.
The parameter koff,Sp1 represents the rate constant for Sp1 ZF-DBD’s spontaneous dissociation from the overlapping target site (Fig. S6). The ratio kdisp/koff,Sp1 corresponds to enhancement of Sp1 ZF-DBD’s dissociation by Egr-1 ZF-DBD.
This corresponds to the second-order rate constant for the overall process at relatively low concentrations of Egr-1 ZF-DBD (i.e., Ptot << Aen).
