Analysis of VZV BAC genome structures. SapI restriction sites flanking the ORF30 gene and the resulting predicted fragment sizes are shown. Insertion of galK results in an additional SapI site. SapI-digested BAC DNAs were visualized after electrophoresis on a 0.65% agarose gel and EtBr staining. Mutants containing alanine substitutions are predicted to be identical to parental VZV_LUC. All mutations were confirmed by sequence analysis. White arrows and dots mark the approximate locations of fragments in Δ30L, Δ30M, and Δ30S.
