Phosphorylation is required for cofilin to interact with the MV N protein. (A) HeLa/hSLAM cells were infected with MV at an MOI of 0.1. Phosphorylated cofilin (p-Cofilin) and total cofilin were detected by immunoblotting at indicated time points. (B) HeLa/hSLAM cells were transfected with expression plasmids encoding GST-N, HA-P, and Flag-Cof or Flag-CofS3A. At 24 h after transfection, the GST-N protein (green) and Flag-Cof or Flag-CofS3A (red) were immunostained. (C) A Co-IP assay was performed with lysates from MV-infected HEK293/hSLAM cells expressing GST-N or HA-P plus Flag-Cof or Flag-CofS3A. At 48 h after transfection, cell lysates were prepared, and proteins were precipitated and examined by Western blotting.
