Synthetic route to a membrane-spanning phosphoethanolamine (MSPE) derivative. Caldarchaeol was converted into caldarchaeo-bis-phosphoric ester dichloride using phosphoroxychloride in the presence of triethylamine. Treatment with ethanolamine produced caldarchaeo-bis-oxazaphospholane, which upon mild acid hydrolysis yielded MSPE as a precursor for membrane-spanning spacer-linked fluorophores (MSPE derivatives) as shown in Fig. 5A.
