The raw data displayed in the upper panel were integrated in the lower panel. The data points represent the experimental injection heat and the solid line corresponds to the calculated fit of the data using a model with two sets of binding sites of the ORIGIN software package (MicroCal Inc.). The heat data of calcium into the reaction buffer was subtracted from the reaction heat data between CabA and calcium. The experimental parameters were as follows: 70 injections of 3 μl of calcium solution, stirring speed of 307 rpm, duration 12 s, and spacing 240 s.
