Initial activation of Gαi/o-coupled receptors leads to a decrease in cAMP production, however, persistent activation leads to heterologous sensitization and a subsequent enhancement in cAMP production that is readily observed by removing the Gαi/o agonist and stimulating the adenylyl cyclase.
