QD fluorescence consists of fluorescent and non-fluorescent states, called on and off states, respectively. We defined the fluorescence intensity below 10% of the maximum fluorescence intensity for each QD particle as its off state. Black boxes indicate the off state. When we measured the fluorescence of fresh QD particles and analyzed their properties, the results indicated that the mean time in the off state during 20 s of observation was approximately 4 s, with a low s.e.m. Therefore, based on an off-state time of 4 s, we selected a single-particle QD using each subtracted image and video image and then measured the fluorescence intensity of the single QD particle.
