(A–B) Ectopic expression of PPM1D decreases the ratio of p-RBM38 vs. RBM38. HCT116 (A) or MCF7 (B) cells were transiently transfected with various amounts of a pcDNA3 vector expressing wild-type PPM1D or phosphatase dead mutant D314A. 24 h post transfection, cells were induced to express RBM38 for 12h. Cell lysates were harvested for Western blot analysis with antibodies against RBM38, PPM1D or actin. (C) PPM1D directly dephosphorylates RBM38 as measured by in vitro phosphatase assay. RBM38 immunocomplexes from HCT116 cells were treated with various amount of GST, GST-PPM1D, or GST-PPM1D-D314A in the presence or absence of Mg2+ or a PPM1D inhibitor as indicated. The ratio of p-RBM38 vs. RBM38 was measured by Western blot analysis with antibodies against RBM38.
