Primary microglia cells from WT (A) and ABCA7 KO (B) mice were used to examine subcellular compartmentalization of APP and endosomes. After 5 min of APP endocytosis, more APP (red) overlapping with endosomes labeled with EEA1 antibodies (green) was observed in ABCA7 KO cells. The scale bars represent 10 μm. The number of subcellular endosomes overlapping with APP after 5min of endocytosis was calculated and normalized with the total number of endosomes (C). Endosomes overlapping with APPs and APPs overlapping with endosomes were significantly increased in ABCA7 KO cells as compared with wild-type cells.
