Detection and localization of SrtB. A, immunoblotting with anti-SrtB detected a major polypeptide of ∼25 kDa in whole cell extracts of the Ptet-srtB strain grown in the presence of the inducer ATc (100 and 500 ng/ml) but not in extracts of wild-type strain 630 (WT) or not-induced Ptet-srtB strain (0 ng/ml ATc). B, Ptet-srtB culture induced with 100 ng/ml ATc was fractionated into cell wall (CW), membrane (Mb), and cytosolic (Cy) compartments and immunoblotted with anti-SrtB polyclonal antibodies. Proteins were separated on 12% SDS-polyacrylamide gels.
