Enhancing PIP2 in the inner membrane leaflet leads to a fortification of the membrane-cytoskeleton shell mediated through ezrin. A redistribution of the actin cytoskeleton occurs in response to an increased PIP2 level (a). Ezrin depletion via ezrin siRNA, neomycin microinjection or ezrin inhibitor NSC 668394, however, results in more condensed, flattened cells (c).
