OSI-027 induces human pancreatic cancer cell apoptosis in vitro. Human pancreatic cancer lines (PANC-1 or MIA PaCa-2) were either left untreated (“C”), or stimulated with applied concentration of OSI-027, cells were then cultured for an indicated time, and cell apoptosis was analyzed by staining assay (A, B) and caspase-3 activity assay (C, D). PANC-1 or primary human pancreatic cancer cells, pretreated with the caspase-3 inhibitor z-DVED-fmk (ZDVED, 30 μM), the caspase-8 inhibitor z-ITED-fmk (ITED, 30 μM), or the pan caspase inhibitor z-VAD-fmk (ZVAD, 30 μM) for 1 h, were stimulated with OSI-027 (100 nM) for 72 h, cell survival was analyzed by CellTiter-Glo assay (E, F).
