NR1 gene silencing in Schwann cells blocks cell migration in response to LRP1 ligands. (A) Images of Schwann cells, transfected with NTC or NR1-specific siRNA, which migrated to the underside surface of Transwell membranes. The cells were treated with EI-tPA (12 nm), α2M* (10 nM) or vehicle (PBS). Migration was allowed to proceed for 4 h. (B) Quantification of cell migration results. Data are expressed as the mean±s.e.m. (n=6, *P<0.01 compared with the vehicle control). (C) Schwann cell death was measured using the cell death ELISA in cells transfected with NTC siRNA or NR1-specific siRNA, after culturing for 4 h in complete medium (10% FBS, CM), Sato medium, 0.5% FBS-containing medium (LSM), or LSM plus TNFα (50 ng/ml). The mean±s.e.m. is shown (n=4, *P<0.05).
