(b) ARS assays for oriP1-cdc6L and oriP2-cdc6M. The plasmids pBI501-oriP1-cdc6L and pBI501-oriP2-cdc6M (Supplementary Table 1) were transferred into strains with the corresponding fragments deleted. Southern blot analysis was performed, using the bla gene as the probe. Lanes P1 and P2 contain plasmids isolated from E. coli with pBI501-oriP1-cdc6L for P1 and pBI501-oriP2-cdc6M for P2, whereas the other lanes contain crude DNA extracted from the corresponding H. mediterranei transformants.
