Ethanol and 4-NQO induce changes in histone acetylation and methylation at transcriptional start sites of some genes involved in ethanol metabolism. Mouse tongue tissue was homogenized, and pre-cleared lysates (15 μg of DNA) were immunoprecipitated (IP'ed) using 0.5 μg of antibodies specific for H3K27ac, H3K27me3, or IgG (negative control). Purified DNA was used for qPCR analysis using primers specific for transcriptional start site (TSS) regions (Table 1) of the (A) Aldh2, (B) Aldh1a3, (C) Adh1, (D) Adh7, (E) Cyp2a5, or (F), Cyp2e1 genes. Binding is expressed relative to the pre-IP input DNA. Error bars represent standard errors of independent mouse tongue samples where n≥5.
