UTX decreases Ezh2/Suz12 binding to the UCP1 and PGC1α promoters. Inactivation of UTX enhances Ezh2 (A) and Suz12 (B) bindings at the UCP1 and PGC1α promoters. Overexpression of UTX decreases Ezh2 (C) and Suz12 (D) bindings at the UCP1 and PGC1α promoters. UTX knockdown or overexpression in BAT1 adipocytes was conducted as described under “Experimental Procedures.” The Ezh2 or Suz12 binding to the UCP1 and PGC1α promoters was assessed by ChIP assays followed by SYBR Green quantitative PCR as described under “Experimental Procedures.” All data are expressed as mean ± S.E., n = 4; groups labeled with different letters are statistically different from each other.
