Strains were induced to sporulate on solid media for 14 hrs then resuspended in liquid sporulation media and incubated with alkyne D-alanine (alkDala). After fixation, permeabilization, and copper-catalyzed cycloaddition of an azide-conjugated fluorophore, Hoechst nucleoid dye was added, and cells were visualized by light microscopy.
