WT and point-mutated AAH effects on Notch signaling genes: Huh7 cells transfected with WT or point-mutated N-Myc-AAH cDNAs were used in duplex qRT-PCR assays to measure (a) Notch-1, (b) Jagged-1, (c) HES-1, and (d) HEY-1 gene expression in which results were normalized to HPRT measured simultaneously in the same reactions. EV-transfected cells served as a negative control, while M19-H→Q-transfected cells served as a positive control for inhibiting Notch signaling due to loss of catalytic activity. Graphs depict the relative mean (± S.E.M) levels of target gene expression.
