Depletion of CD14+ macrophages leads to a marked decrease in the ability of MMC to produce TNF and IL-6, both constitutively and in response to LPS stimulation. CD14+ macrophages were depleted from MMC isolated from the colon of ART-naïve AIDS patients. Total (CD14+) and CD14-depleted MMC were then cultured for 18 h in the presence or absence of LPS. Cytokine/chemokine secretion into the culture supernatant was quantified using customized Bio-Plex plates. Results represent the mean ± SD of triplicate assays performed on 10 African patients with AIDS. Statistical significance between groups was calculated using the Mann–Whitney U test.
