(A) Schematic of Chr3ΔR constructs. The right arm of Chromosome III (Chr. 3) is deleted in both constructs. Chr3ΔR-12ARS contains 12 ARSs on the remaining region, while Chr3ΔR-7ARS contains 7 ARSs.
(B) PFGE and FACS analyses of cells containing Chr3ΔR. Experiments were similar to Figure 5C. A representative result and quantification of 6 trials are shown with means and SD plotted; the values were normalized to wild-type cells containing Chr3ΔR-12ARS in both plots. The left plot shows that Chr. 3 is replicated to a lesser extent in rtt107Δ cells than in wild-type cells. The right plot shows that Chr3ΔR-7ARS replicated more poorly than Chr3ΔR-12ARS in rtt107Δ cells, but not in wild-type cells. FACS shows that the two versions of Chr3ΔR did not affect S phase progression.
