K442 is dispensable for nuclear rim delivery and propagation in epithelial cells. (A) The number of fluorescent capsids at nuclear rims was estimated 2 hpi in PK15 cells. Representative images document that while the majority of input viral particles remain in proximity of cell surfaces, a fraction have traversed the cytoplasm and are now localized at the nuclear periphery. Mean ± SD of capsid fluorescence at nuclear rims are shown with statistics based on an unpaired t test. (Scale bar: 10 μm.) (B) Propagation kinetics of the K442R mutant on PK15 cells (MOI 10) relative to WT and repaired PRV. PFU, plaque-forming units.
