mCherry was fused in-frame between pUL25 amino acids 42–43 for PRV and 50–51 for HSV-1.

Amino acid numbers are based on PRV strain Becker (GenBank JF797219) and HSV-1 strain F (GenBank GU734771 and KM222724).

Soluble GFP cassette driven by a CMV promoter was inserted into the US4 gene. This virus is also deleted for the UL25 gene.
