Speculative working model for the mutual regulation between Tpr and Aurora A in mitotic progression and centrosome homeostasis. We propose that the coiled-coil Tpr-M domain binds to and sequesters endogenous and exogenous Aurora A and thereby causes a reduction in Aurora A phosphorylation. This action is analogous to that of the Aurora A inhibitor, Alisertib. This domain merits further investigation as a therapeutic agent for those cancers in which Aurora kinaseA is over-expressed.
