Bisulfite sequencing analysis for the methylation status of the 98 bp region of the GFPint98(+) transgene. DNA was extracted from non-infected (upper panel) and PSTVd-infected (crossed with Nt-Nbdi) (lower panel) Nt-GFPint98(+) plants and subjected to bisulfite sequencing. In each case, 10–15 clones were sequenced and data were interpreted with CyMate software.88 ‘Ref’ corresponds to the reference sequence (not bisulfite treated).
