The non-PCR amplified sample (COLO 829 DC) is purely homoplasmic reference (G), but the LR sample shows G/A heteroplasmy. Sequencing traces for the COLO 829 LR (B) and COLO 829 gDNA(C) confirm that the PCR step did induce the false heteroplasmy. Position 3850 is indicated with the rectangle and arrow in both panels, showing heteroplasmy (black and green peaks colocalizing) in the LR sample.
