A: The optical density of MTT is often higher in the cells at 39°C than at 37°C, even when fewer cells at 39°C are observed under microscope, indicating that cell viability may be overestimated at 39°C when MTT assay is used. B, C and D: Treatment of Hela, A549 and HCT116 cells with 1 μM cisplatin (CDDP), 4 μM 5-FU or 0.05% clove bud ethanol extract significantly decreases the cell viability compared with the untreated control at the same temperature (a in C, B and D, p<0.05; t test), whereas comparison between 37°C and 39°C is not recommended.
