A) Chemical structures of α and βGalcer. B) Localization of endogenous αGalcer in RBL-CD1d cells. RBL-CD1d cells were fixed and permeabilized with saponin before staining with antigen-purified rabbit anti-αGalcer antibody (red) and anti-LBPA antibody (green). Both LBPA, a constituent of the lysosomal membrane, and αGalcer localize to intra-cellular vesicles and demonstrate some overlap (white arrowheads). Immunofluorescence images were collected using Confocal microscopy. Flattened Z series are presented, and scale bar is indicated in the image.
