HeLa cells were untreated (Un) or treated with the various concentrations of purified SLO that are shown on the top of Panels A and B. The presence of PAR analyzed by Western blotting as shown, following 5 (A) or 1 (B) hours post-treatment. Formation of PAR was also assessed in HeLa cells infected with S. pyogenes strains expressing monomer or pre-pore locked mutant SLO proteins with NADase+ (Mono or Pre) or NADase (MonoSPN or PreSPN) SPN and compared with uninfected (uninfect.) cells and cells infected with strains expressing wild-type SLO, but lacking SPN (ΔSPN) or expressing NADase SPN (NADase). Formation of PAR was assessed by Western blotting at 5 hours post-infection, as shown (C). Migration of molecular weight standards are shown on the left.
