NbSOBIR1 and NbSERK3 (BAK1) are required for LepR3-mediated development of cell death. (A) 2-week-old N. benthamiana seedlings were subjected to virus-induced gene silencing (VIGS) by inoculation with the TRV:NbSOBIR1, TRV: NbSERK3(BAK1), and TRV:GFP, respectively. TRV:GFP served as a control. Three weeks after TRV inoculation, PR1a-AvrLm1/LepR3, PR1a-AvrLm1/GFP, and LepR3/GFP were transiently co-expressed in the order indicated on the image. Cell death caused by Transiently expressed BAX served as a control. Leaves were photographed 6 days after infiltration. The experiment was repeated three times with 10 plants for each TRV construct. Numbers below the panels indicate the occurrence or the absence of HR within 30 sites co-infiltrated with PR1a-AvrLm1/LepR3. (B) qRT-PCR analysis showing NbSOBIR1 expression in N. benthamiana inoculated with the indicated TRV VIGS constructs (TRV:GFP and TRV:NbSOBIR1) and in wildtype (WT) N. benthamiana without TRV inoculation.
