A. MDA-MB-231-TGL cells were transduced with control lentivirus (pLKO.1) or three different shRNA constructs to knock-down RhoE expression. Knock-down efficiency was analyzed by Western blotting. Numbers show the relative expression level of RhoE after quantification by densitometry. B. After injection of MDA-MB-231-TGL cells (control and RhoE knock-down, using two different shRNAs) in the tail vein of 6 nude mice (2 per construct), lung tumors were analyzed every week by in vivo bioluminescence imaging and normalized photon flux was plotted (left graph). The image on the right shows representative results of mice injected with control (pLKO.1) and shRhoE #3 MDA-MB-231-TGL cells. C. Lung colonization by MDA-MB-231 cells. At the end of the experiment, lungs were removed and inspected to confirm the presence of tumors resulting from the injection of control (pLKO.1, left) and RhoE knocked-down (shRhoE #3, right) MDA-MB-231-TGL cells. Hematoxylin and eosin staining (bottom images, 200x) revealed no differences between control- and shRhoE- induced metastases tumors.
