Semi-quantitative RT-PCR of GPCR targets in Tetrahymena transformed with siRNA. Tetrahymena were transformed with siRNA and then DNA-free RNA was isolated and subjected to an RT-PCR assay that semi-quantitates transcript levels based on the number of PCR cycles required for amplicon visualization using agarose gel electrophoresis (Carlson et al., 2007).
