PknB_PASTA reduces growth of Mycobacterium tuberculosis. (a) Overexpression of PknB_PASTA domain delays growth initiation. Mycobacteria (approx. 1 × 10⁶) were inoculated in supplemented 7H9 medium containing hygromycin and tetrocycline. (b) Effect of different concentrations of rPASTA on M. tuberculosis growth. Mycobacteria (10³ cells ml⁻¹) were inoculated in supplemented 7H9 medium containing different concentration of sterile rPASTA. The optical density was measured after 21 days of incubation. (c) Regrowth of stored M. tuberculosis was delayed by the addition of rPASTA. M. tuberculosis (2 × 10⁴ cells ml⁻¹) was inoculated in supplemented 7H9 medium. Buffer was used as control and rPASTA was added at final concentration of 10 µg ml⁻¹. (b,c) Each data point is an average value of 3 biological replicates, error bars indicate standard deviations. Experiments were performed three times and results of one typical experiment are shown.
