d-Ala deconvolution and contributions in S. aureus and E. faecium. (a,b) Complete accounting of whole-cell d-alanine is possible by experimental deconvolution using [15N]Gly/d-[1-13C]Ala-labelled whole cells. d-Ala contributions from peptide stems are analytically determined by spectral selection. By difference, the remaining d-Ala contributions are attributed to teichoic acid d-Ala. (c) Direct comparison of the 13C NMR spectrum of whole cells with isolated cell walls in which esterified d-Ala is lost from teichoic acid confirms the identification of teichoic acid d-Ala.
