TLC measurement of phospholipids in wild-type (WT) and FAB1A-GFP-expressing plants. Five-day-old seedlings were labeled overnight with 32Pi, after which they were further incubated for 2 h in the presence (YM) or absence (DMSO) of 1 μM YM201636. After extraction, lipids were separated by TLC and quantified by phosphoimaging. Data are expressed as the percentage of total 32P-phospholipids. The levels of PtdIns(3,5)P2 (A), PtdIns(4,5)P2 (B), PtdIns3P (C), phosphatidic acid (PA; D), phosphatidylinositol (PI), phosphatidylcholine (PC), phosphatidylethanolamine (PE), and phosphatidylglycerol (PG; E) are shown. Three independent samples containing three seedlings each were used. *, Significant differences between seedlings treated with or without YM201636 are indicated by (Student’s t test, P < 0.001).
